چکیده

Kobayashi, N; Procop, GW; Fujishiro, T; Tuohy, MJ; +Bauer, TW +The Cleveland Clinic Foundation, Cleveland OH [email protected] Introduction The importance of bacterial biofilm associated with joint implants has been recognized widely and investigated. These bacteria are best detected after the biofilm has been disrupted by, for example, ultrasonication. Although ultrasonication enhances detection of adherent bacteria, it may kill the organisms, decreasing the sensitivity of conventional culture results. In addition, the optimum duration of ultrasonication to detect biofilm formative bacteria has not been investigated. The aim of this study was to investigate the effect of ultrasonication on the viability of adherent bacteria, to compare the use of polymerase chain reaction (PCR) and culture to detect organisms after ultrasonication, and to determine the optimum duration of ultrasonication to maximize detection of bacteria adherent to a metal substrate.

Comparison of OmpA Gene-Targeted Real-Time PCR with the Conventional Culture Method for Detection of Acinetobacter baumanii in Pneumonic BALB/c Mice

Background: Acinetobacter baumannii is an important pathogen in health care and is responsible for severe nosocomial and community-acquired pneumonia. To design novel therapeutic agents, a mouse model for A. baumannii pneumonia is essential. Methods: We described a mouse model of A. baumannii using clinical and 19606R standard strains for developing a quantitative real-time PCR (qRT-PCR) for ra...

Molecular study of biofilm gene of sulfate reducing bacteria (SRB) isolated from patients with periodontitis and the effect of aloe vera plant extract on its expression by Real time-PCR method

Background and Aims: Due to the increasing problems and side effects of the use of chemical antibacterial agents as well as antibiotic resistance, this study aimed to evaluate the effects of aloe vera gel on biofilm gene expression of sulfate-reducing bacteria (SRB) isolated from patients with periodontal infection by Real time-PCR method. Materials and Methods: For this study, 100 individu...

Real-time Pcr without Dna Extraction: a Modification for More Rapid Detection of Bacteria on Orthopaedic Implants

Introduction Infection is one of the most important causes of joint implant failure. The polymerase chain reaction (PCR) is a sensitive way to test for bacteria, but most PCR procedures take too long to provide results during a revision arthroplasty operation. For example, we have described the sensitivity and specificity of real-time PCR combined with ultrasonication of retrieved implants to d...

Rapid quantitative detection of Listeria monocytogenes in chicken using direct and combined enrichment/qPCR method

Listeria monocytogenes is a species of foodborne pathogen often related to foods, such as poultry, ready-to-eat products, fruits, and vegetables. The culture method is a standard procedure for the detection of bacteria in food products. The real-time quantitative PCR (qPCR) technique can be used for the quantification of foodborne pathogens. The current research was aimed to assess...

Molecular study of Candida albicans isolated from periodontal infections and effect of Sargasum alga extract on biofilm als gene expression using Real-Time-PCR

Background and Aims: Candida is an opportunistic pathogen that causes illness in people with a defective or weakened condition. Infectious diseases (periodontal diseases) are inflammatory and malignant inflammation of the dental-gum complex, in which the growth of biofilms caused by Candida glabrata, Parapseilosis and Tropikalis is less than Candida albicans. Brown algae Sargasum is more compat...
